VALDOXAN (Agomelatine) is a new antidepressant with selective agonist actions at melatonin receptors (MT1 and MT2 ) and selective antagonist action at serotonin 5HT-2C receptors ,it blocks serotonin 2C (5-HT2C) receptors.

This results in the release of norepinephrine and dopamine (the drug does not affect serotonin levels.

It does not affect the uptake of serotonin,nor adrenaline or dopamine Agomelatine increases nor adrenaline and dopamine release specifically in the frontal cortex and has no influence on the extra cellular levels of serotonin(compared to SSRIs, SNRIs and the older tricyclic antidepressants)

The melatonergic function appears to improve sleep patterns, whereas the serotonergic antagonism results in the release of norepinephrine and dopamine. Given the current information, the overall side-effect profile of agomelatine appears relatively mild. For example, agomelatine has no discontinuation syndrome, exhibits infrequent sexual dysfunction, and is generally weight neutral

Treatment with the drug increased slow wave sleep without modification of REM (Rapid Eye Movement) sleep amount or REM latency. Agomelatine also induced an advance of the time of sleep onset and of minimum heart rate. From the first week of treatment, onset of sleep and the quality of sleep were significantly improved without daytime clumsiness as assessed by patients.

Agomelatine appears to be well tolerated, have low rates of sexual dysfunction, exhibit no discontinuation syndrome, and be weight neutral. Agomelatine improves sleep and, at present, seems to be relatively safe in overdose.

However, this antidepressant may cause elevations in hepatic enzymes,a finding that warrants further investigation so the doctor may order liver tests prior to treatmnet with it and further follow ups after .

Effexor (venlafaxine) is an antidepressant in a group of drugs called selective serotonin and norepinephrine reuptake inhibitors (SSNRIs). Venlafaxine affects chemicals in the brain that may become unbalanced and cause depression.

>Agomelatine appears to cause fewer sexual side effects and discontinuation effects than paroxetine. It appears better tolerated than the SSRI

As a conclusion ,check with your doctor for what is best for you ,in general valdoxon is anew drug treatment for major depression disorder affects receptors without affecting central receptors and serotonin uptake ,it increases noradrenaline and dopamin in the frontal cortex and had good sleep issues in comparison to other ssnri one of them being effexor.
